ABI 7900HT, 7500, ViiA 7 Real-Time Quantitative PCR Systems

Two ABI PRISM® 7500 Sequence Detection Systems, an ABI PRISM® 7900HT Sequence Detection System, and a ViiA 7 Real-Time PCR System (TaqMan, Applied Biosystems) are available in the DBMR. Depending on the machine, the TaqMan machines allow quantification of DNA sequences (e.g. cDNA, genomic DNA) in either individual reaction tubes, array cards, 96-well, or 384-well microtiter plates (reaction volume 20-50 microlitres).

The ABI PRISM® 7900HT System can also process microfluidic cards (MFCs) which consist of 384 individual reaction wells (1 ml per well). MFCs can be customized by the user and the desired primer/probes are applied to the wells during fabrication. Processing is extremely simple: DNA is loaded into fill ports and distributed via micro channels into 47 or 95 reaction wells by centrifugation. After sealing, the cards are ready for quantitative real-time PCR. MFCs have to be ordered in sets of 10 from Applied Biosystems.

On the ViiA 7 Real-Time PCR System, heating blocks can easily be changed from one plate format to another.

  • The ViiA 7 Real-Time PCR System is located in the MEM-Building, Murtenstrasse 35, room G 818

All TaqMan machines are available to all users on a first come, first served basis, priority being given to groups from the DBMR. Users can reserve time slots for their experiments by signing-up in the reservation system. Please note that members of the DBMR can make reservations for up to 1 week in advance. Only ONE RUN per day and user.
Non-DBMR users are requested to make reservations no longer than 3 days before the experiment.
Wednesday afternoon and Friday, the ABI 7900 is reserved for MFC (microfluidic cards), the other days, primarily for normal qPCR at the following time schedule: 08:00-10:30 h, 10:30-13:00 h, 13:00-15:30 h, 15:30-18:00 h. Only authorized people are allowed to change the system (except for ViiA 7). Please note that data from the different systems are not directly compatible.
Everyone should keep strictly to the daily time schedule to prevent delays for subsequent users.
