Abstract In the last decades genetically modified mice have become increasingly important in fundamental and translational research. They have contributed to major breakthroughs in our understanding of the molecular basis of cancer and other diseases. Continuous technology development has resulted in a broad spectrum of different types of genetic modifications. These range from conventional transgenes and knockouts to tissue specific and inducible overexpression or inactivation combined with ‘in vivo’ reporting systems. Examples will be given in this presentation. The Animal Modeling Facility of Netherlands Cancer Institute has a leading position in generating genetically engineered mouse (GEM) models and supports research groups in developing project accustomed models. This support starts with thorough discussions with the researches what modification are required for answering their scientific questions. This will define what strategy will be taken to generate the proper genetically modified mouse strain. Subsequently, the facility will perform all the necessary molecular, embryological and animal care procedures leading to the desired GEM strain.
